Variants in brain-related genes with a low NSS score confer significantly higher susceptibility than variants in other brain-related genes. The enrichment is strongest for schizophrenia, but we cannot rule out enrichment for other phenotypes. The false discovery rate conditional on the evolutionary proxy points to 27 candidate schizophrenia susceptibility loci, 12 of which are associated with schizophrenia and other psychiatric disorders or linked to brain development. Conclusions: Our results suggest that there is a polygenic overlap between schizophrenia and NSS score, a marker of human evolution, which is in line with the hypothesis that the persistence of schizophrenia is related to the evolutionary process of becoming human.]]> Wed 12 Aug 2020 09:42:59 AEST ]]> Determinants of suicidal ideation and suicide attempts: parallel cross-sectional analyses examining geographical location https://ogma.newcastle.edu.au/vital/access/ /manager/Repository/uon:16783 Wed 11 Apr 2018 17:18:32 AEST ]]> Leveraging genomic annotations and pleiotropic enrichment for improved replication rates in schizophrenia GWAS https://ogma.newcastle.edu.au/vital/access/ /manager/Repository/uon:26719 -8). Predictor variables included demographic factors, pre-dispositional factors, recent difficulties and support, mental health, rural exposure and district-level contextual factors. Results: Gender, age, marital status, and personality made the largest contribution to at-risk alcohol use. Five or more adverse life events in the past 12 months were also independently associated with at-risk alcohol use (Adjusted Odds Ratio [AOR] 3.3, 99%CI 1.2, 8.9). When these individual-level factors were controlled for, at-risk alcohol use was associated with having spent a lower proportion of time living in a rural district (AOR 1.7, 99%CI 1.3, 2.9). Higher alcohol consumption per month was associated with higher district-level socio-economic ranking, indicating less disadvantage (AOR 1.2, 99%CI 1.02, 1.4). Rural exposure and district-level contextual factors were not significantly associated with lifetime consequences of alcohol use. Conclusions: Although recent attention has been directed towards the potential adverse health effects of district or community level adversity across rural regions, our study found relatively few district-level factors contributing to at-risk alcohol consumption after controlling for individual-level factors. Population-based prevention strategies may be most beneficial in rural areas with a higher socio-economic ranking, while individual attention should be focused towards rural residents with multiple recent adverse life events, and people who have spent less time residing in a rural area.]]> Tue 08 Jan 2019 15:22:11 AEDT ]]> Evaluating the efficacy of an integrated motivational interviewing and multi-modal exercise intervention for youth with major depression: Healthy Body, Healthy Mind randomised controlled trial protocol https://ogma.newcastle.edu.au/vital/access/ /manager/Repository/uon:44202 Tue 06 Jun 2023 14:36:21 AEST ]]> Cross-disorder analysis of schizophrenia and 19 immune-mediated diseases identifies shared genetic risk https://ogma.newcastle.edu.au/vital/access/ /manager/Repository/uon:46890 Tue 06 Dec 2022 12:02:52 AEDT ]]> Contribution of copy number variants to schizophrenia from a genome-wide study of 41,321 subjects https://ogma.newcastle.edu.au/vital/access/ /manager/Repository/uon:34283 −15), which persisted after excluding loci implicated in previous studies (OR = 1.07, P = 1.7 × 10−6). Additional analyses revealed a constant floor effect in the amount of variance explained, consistent with the omnigenic model. Thus, we report that putative core gene sets showed a significant effect above and beyond the floor effect that might be linked with the underlying omnigenic background.
